چکیده

A model of arthritis was established by the injection of type II collagen into mice. Only mice bearing the H-2q haplotype were susceptible to the disease. Susceptibility was further mapped by the use of recombinant strains on the Iq locus. Type II collagen arthritis was observed in the (resistant X susceptible) F1 cross. Mice strains were designated high, intermediate, or low responders with respect to the anti-type II antibody levels measured by radioimmunoassay. Arthritis-susceptible strains were all classified as high antibody responders. The clinical and histological appearance of type II collagen arthritis in the mouse indicates that it may be a good animal model for the investigation of various immunogenetic traits in rheumatoid arthritis.

Type Ii Collagen - Induced Arthritis in Mice

The induction of immunological reactivity to type II collagen, a major component of cartilage, has recently been used to establish an experimental model of arthritis in the rat (1). Trentham and colleagues (1) have shown that the intradermal injection of collagen in Freund's complete or incomplete adjuvant elicits an arthritogenic reaction, specifically with native type II collagen. Arthritis o...

HLA-DQ8 transgenic mice are highly susceptible to collagen-induced arthritis: a novel model for human polyarthritis

Genetic studies have indicated that susceptibility to rheumatoid arthritis (RA) maps to the HLA-DR locus of the major histocompatibility complex. Strong linkage disequilibrium between certain HLA-DQ genes and HLA-DR genes associated with RA, however, suggests that HLA-DQ molecules may also play a role in RA susceptibility. To examine the role of HLA-DQ molecules in arthritis, we generated trans...

Resistance to collagen-induced arthritis in a nonhuman primate species maps to the major histocompatibility complex class I region

Type II collagen-induced arthritis (CIA) is an experimentally inducible autoimmune disorder that is, just like several forms of human arthritis, influenced by a genetic background. Immunization of young rhesus monkeys (Macaca mulatta) with type II collagen (CII) induced CIA in about 70% of the animals. One major histocompatibility complex (MHC) class I allele was present only in young animals r...

Genetic control of antibody production during collagen-induced arthritis development in heterogeneous stock mice.

OBJECTIVE To identify genetic factors driving pathogenic autoantibody formation in collagen-induced arthritis (CIA), a mouse model of rheumatoid arthritis (RA), in order to better understand the etiology of RA and identify possible new avenues for therapeutic intervention. METHODS We performed a genome-wide analysis of quantitative trait loci controlling autoantibody to type II collagen (anti...

Collagen epitope expression on B cells is sufficient to confer tolerance to collagen-induced arthritis

BACKGROUND The mechanisms underlying tolerance induction and maintenance in autoimmune arthritis remain elusive. In a mouse model of rheumatoid arthritis, collagen type II (CII)-induced arthritis, we explore the contribution of B cells to antigen-specific tolerance. METHODS To generate expression of the CII-peptide specifically on B-cell major histocompatibility complex type II, lentiviral-ba...
